There can be substantial inter-laboratory variations in the levels of DNA damage measured by the comet assay in different laboratories. Researchers from 14 different laboratories across Europe took part in an inter-laboratory variation in DNA damage using a standard comet assay protocol.
We were pleased to see that the majority of the labs were using Comet Assay IV for scoring their comet samples. The report concluded that the biggest areas for variation between the laboratories were within the experimental procedure (e.g. electrophoresis, gel substrate, different equipment). However, further attempts to reduce variation may focus on the analysis of images in the software systems. This study indicated that future comet assay validation trials should take steps to evaluate the implementation of standard procedures in participating laboratories.
